**Role Responsibilities** High throughput (HTP) protein purification scientist. Responsibilities include operating automated protein production and liquid handling equipment, conducting high throughput, small-scale protein purification and analysis, managing protein inventories, and maintaining a database to support departmental needs for antibody production and characterization. Work directly with other scientists focusing on high throughput molecular cloning and mammalian cell transfection/expression. Specific responsibilities include: Manage protein purification and characterization for hundreds of therapeutic candidates. Develop, optimize and perform small-scale, robotic purification of proteins using Protein A chromatography. Develop, optimize and perform small-scale, robotic buffer exchange purification on weekly basis. Develop, optimize and perform small-scale robotic protein characterization. Maintain and run high pressure liquid chromatography (HPLC) instruments. Perform robotic sample distribution to end users, including transfer to multiple small vials and adjustment of protein concentration. Upload purification and characterization data into departmental sample database daily. Develop and manage sample/request database. Interface with requestor for sample and data transfer. Perform automated protein assays (enzyme linked immunosorbent assay - ELISA or equivalent). Manage protein purification laboratory: Prepare and maintain inventories of protein purification chromatography apparatus. Represent high-throughput protein production group at multiple project team meetings, present data and report back on planned resource needs from teams. Perform high-throughput mammalian cell transfection and expression experiments. Conduct all cell-handling and purification work with good sterile technique. Perform automated, small-scale DNA purification. Conduct basic microbiology procedures (DNA transformation, purification, single cell isolation). Prepare DNA samples for sequencing; perform sequence analysis. Conduct basic molecular biology procedures (enzymatic DNA digestion, DNA purification, gene cloning by ligation or isothermal assembly, etc). Manage other laboratory scientists performing assays and small scale protein production.

**Basic Qualifications**

Educational Requirements:

Bachelor's Degree in Biochemistry, Biology, Biotechnology or related field and 5 years' experience in the job offered or related. Or a Master's in Biochemistry, Biology, Biotechnology or related field and 3 years' experience in the job offered or related.

**Preferred Qualifications**

Special Skills Requirements:

(1) At least 3 years of experience with automated small scale protein purification technologies including high pressure liquid chromatography (HPLC; e.g. Agilent) system operation, maintenance and data analysis (2) 2 years of experience with transient mammalian cell culture (transfection/ expression) including HEK293/ CHO systems.

(3) At least 3 years of experience with Tecan FreedomEvo and BeckmanCoulter Biomek liquid handling instrumentation and software including method development, operation and maintenance.

(4) At least 3 years of experience with high throughput assay development using automation (e.g. ELISA, HTRF, flow cytometry) including data analysis and presentation of large data sets using TIBCO Spotfire or similar software.

(5) Experience developing and running non-specificity assays for protein therapeutics.

(6) Experience with antibody discovery technologies.
