The Bacterial Vaccines & Technology department within Pfizer is focused on the development of vaccines to prevent infectious disease. Though we specialize in bacterial vaccine development, the group also contributes to viral vaccine programs through technologies such as genetic engineering and antibody discovery . The task of the B-cell technology group is the discovery of monoclonal antibodies that recognize infectious disease targets. These antibodies are utilized in applications ranging from validating target vaccine candidates and exploration of human immune responses. Several techniques are used for the generation of these antibodies including synthetic engineering, screening human and/or mouse memory B-cells, and potentially by using yeast and phage libraries.



**How you will participate**

As a Scientist, you will be embedded in our operations and your work will be part of the on-going vaccine programs to deliver first in-class medicines in the upcoming years. You will be an essential member of the B-cell team generating and characterizing different types of antibodies and reagents for vaccine characterization and development of clinical testing assays.

With your deep knowledge you will support the team especially for B-cell based antibody generation using an internally established discovery platform which we continuously improve. Your creativity and experience will be essential for this task. Experience in protein characterization and ligand binding assays is advantageous but not required.

Your ability to plan and coordinate will support the team to achieve short-term projects as well as long-term goals and maintain the reagent inventory in good quality.

Using your prior work experience, scientific judgement to adjust existing methods combined with consulting others will allow to streamline processes.

With your scientific enthusiasm you will support the team to achieve new goals and timelines to help patients.



**Your role responsibilities**



+ Execute generation of monoclonal antibodies by single B-cell cloning with application of various molecular biology methods such as Gibson/In-fusion or restriction enzyme cloning. Screening of clones using ELISA, Luminex, flow cytometry techniques. Contribution to new ideas and initiatives, and participation in project planning activities.

+ Characterization of antibodies by different functional assays such as binding or neutralization assays using ELISA, Octet, Luminex, Biacore or flow cytometry.

+ Establish assays for antibody characterization individually or together with other teams.

+ Analyze, interpret and summarize experimental data. Participate in the evaluation of new methods or technologies.

+ Present experimental results at group meetings and project meetings as required. Write research reports.

+ Experience in antibody/protein purification using AKTA systems is preferred.

+ Maintain laboratory notebook in a detailed and descriptive manner. Keep current entries into the antibody and plasmid databases.

+ Should be capable of multitasking priorities and handle various tasks in parallel while demonstrating problem solving skills in the planning and execution of objectives.

+ Reagent inventory maintenance using databases, e.g. LIMS





**BASIC QUALIFICATIONS**

B.S. in Molecular Biology, Immunology, or a related discipline plus 6 years of relevant experience. Background in working with microbial pathogens and sterile cell culture technique is mandatory.



+ Candidate should be proficient in general molecular biology and immunology techniques,

+ Familiarity with mammalian cell culture as well as expression systems for antibodies, bacterial and viral proteins

+ Experience in various cloning techniques, isolation of nucleic acids, construction of plasmid vectors,

+ Performing PCRs, and nucleotide sequencing is expected.

+ Hands on experience in the following areas is required:

+ Use of DNASTAR Lasergene/Vector NTI software,

+ ELISA or Luminex microplate assays,

+ Protein quantitation (UV/colorimetric) assays and SDS-PAGE/western blot analysis

+ Antibody purification(affinity, IEX, SEC) experience is preferred.

+ Knowledge of yeast and phage display techniques is not required but would be advantageous.

+ Proficiency with Microsoft Office applications and relevant scientific software, including bioinformatics and graphing/statistical analysis tools is expected.

+ The incumbent must possess excellent verbal and written communication skills.





**PREFERRED QUALIFICATIONS**

M.S. degree in Molecular Biology, Immunology, or a related discipline plus 4 years of relevant experience.

Experience in protein characterization and ligand binding assays is advantageous.



Sitting, standing, walking, light lifting (<25> Travel to regional, national and international meetings. Adherence to safe work practices and procedures required.
